In a method for the determination of glutamate oxalacetate transaminase or glutamate pyruvate transaminase by reacting oxaloacetate or pyruvate with glutamate to form α-ketoglutarate in buffered solution, α-ketoglutarate formed is set in the presence of γ-aminobutyrate from γ-aminobutyrate transaminase to form succinate semialdehyde, reduced with the latter NADP in the presence of succinate semialdehyde dehydrogenase to NADPH and measures this either directly or after transfer with a tetrazolium salt and an electron transfer agent, such as, for example Diaphorase, phenantroline methosulfate or phenazine methosulfate in a formazan dye. A reagent suitable for this process contains γ-aminobutyrate transaminase, succinate semialdehyde dehydrogenase, γ-aminobutyrate, glutamate, buffer, either oxaloacetate or pyruvate and optionally also tetrazolium salt, electron transfer agent and surface-active agent.
