PURPOSE:To accomplish mass proliferation of the title virus for the study of e.g. viral infantile diarrheic pathogen by inoculating human adenovirus type 40 into cell strain capable of proliferation of human adenovirus type 40 in human lung adenocarcinomatous cells to make a culture of said virus and to effect mass production of plaque. CONSTITUTION:Human adenovirus type 40 is inoculated into cell strain [e.g. A549 cells (ATCC CCL-185)] capable of proliferation of human adenovirus type 40 in human lung adenocarcinomatous cells and agar containing 25mM of MgCl2 is put on the cells to make a culture of said virus for 10 days. On day 10 after virus infection, the cells are stained with e.g. Neutral Red to confirm that plaque has been formed, and from the plaque, virus is collected and then proliferated in the above-mentioned cell strain. This operation is then repeated several times, thus accomplishing the objective mass production of plaque-purified human adenovirus type 40 as a single virus.
