CARRIER FOR DETERMINATION OF MUTATION OF BASE SEQUENCE OF NUCLEIC ACID AND DETECTION OF NUCLEIC ACID AND REAGENT FOR DETERMINATION OF MUTATION OF NUCLEIC ACID AND DETECTION OF NUCLEIC ACID CONTAINING THE CARRIER
PURPOSE:To enable sharp detection of the subject substance by bonding and immobilizing a single stranded nucleic acid composed of nucleotide to the surface of a particle having a specific diameter. CONSTITUTION:A polymeric compound such as styrene is emulsion-polymerized to obtain particles (A) having non-porous surface, holding hydroxyl groups on the surface, insoluble in an organic solvent to be used in the activation process of the hydroxyl group, insoluble in water and having particle diameter of 0.1-10mum. The component A is activated with tresyl chloride, etc., to obtain an activated carrier (B). Separately, a single stranded nucleic acid probe (C) having a primary amino group bonded to a terminal of a nucleic acid through a spacer such as ethylenediamine and composed of =10 nucleotides is prepared. The component C is added to the surface of the component B and immobilized by the reaction of formula (spacer is straight or branched-chain compound having 2-50 atoms at the skeleton part) to obtain a reagent for the determination of the mutation of nucleic acid base sequence and the detection of nucleic acid.
