PROBLEM TO BE SOLVED: To measure the residual activity of AChE simply, highly accurately, safely, and inexpensively by directly determining acetic acid generated by the action of AChE by ion chromatography with a electric conductivity detector. ;SOLUTION: An insect mixed into a food is extracted and cleaned by pure water, and the head of the insect is cut out to be a sample to be measured. After cleaning, an extraction buffer solution is added to the sample to be measured. The sample is ground down by a glass rod, etc., homogenized by ultrasonic waves as being cooled by ice, and centrifuged to obtain a centrifugation supernatant liquid as a crude enzyme liquid. By determining acetic acid in the crude enzyme liquid obtained in this way, the residual activity of acetylcholine esterase AChE of the insect mixed into the food is measured. Ion chromatography with an electric conductivity detector is most favorable as the determining method of acetic acid in the crude enzyme liquid to obtain highly accurate results.
