PROBLEM TO BE SOLVED: To provide a method for detecting microorganisms, high in detection limit for microorganisms, suitable for quickly assaying many specimens and enabling assay over a wide pH range, and to provide a reagent kit and a system for assaying luminescence.;SOLUTION: The method for assaying the proliferation and/or activity of microorganisms comprises incubating the microorganisms together with an oxidized quinone in a culture solution and making a quantification by reacting the product with a chemoluminescent reagent, wherein the reaction is carried out in the presence of at least one metal ion selected from the group consisting of molybdenum, manganese, nickel and cobalt ions. The reagent kit for assaying luminescence for detecting microorganisms comprises the oxidized quinone, the chemoluminescent reagent and at least one compound as a supply source for the metal ion selected from the group consisting of molybdenum, manganese, nickel and cobalt ions.
