The device has a light source (10) i.e. laser light source, for generating light beam (11), a paraboloid prism (17) and a polarizer (12) for polarizing the light beam. A variable mirror (14) is provided for directing the light beam to an adjustable location of the paraboloid prism such that an incident angle of the light beam on a sample i.e. flow cell (20), is changed through changing of the adjustable location. An immersion layer (18) i.e. oil immersion layer, is arranged between the paraboloid prism and the sample, and two transparent plates are provided for enclosing the sample. An independent claim is also included for a microscoping method.
