Provided in the present invention is a sustained-release oral formulation, comprising a leaf extract of Ginkgo biloba and a sustained-release matrix system. According to the present invention, the sustained-release oral formulation of the leaf extract of Ginkgo biloba is capable of regulating the absorption degree of an active ingredient in vivo by a controllable release speed. The formulation exhibits continuous absorption by the delayed release speed of the formulation while penetrating through a gastrointestinal tract, when being injected in the human body, and continuously exhibits platelet activating factor (PAF) inhibitory activities and antioxidant activities. Therefore, the formulation is expected to improve a patient compliance of dosage by once-per-day dosage, and to have high effects of treatment.
