isolated nucleic acid summary; yeast; method for producing ethanol; strain of h. polymorpha; and a gene identification method are methods for identifying ethanol tolerance genes in yeast, ethanol tolerance genes, and mutant strains used to identify such genes. a gene designated herein as hpett1 has been isolated from hansenula polymorpha yeast. hpett1 expression in an h mutant strain. The ethanol-sensitive polymorpha designated as 7e complemented the mutant's ethanol sensitivity. When multiple copies of hpett1 were integrated into the genome and overexpressed, the transformed strain showed approximately 10-fold higher resistance to ethanol and protein folding agent azc. hpett1 expression also increased ethanol tolerance in saccharomyces cerevisiae. hpett1 has 39% sequence identity with a previously identified protein from s. cerevisiae denoted mpe1, however, the mpe1 gene does not confer ethanol resistance to mutant 7e. another pichia stipites yeast gene has been identified which encodes an orthologous protein that has 37% identity for hpett1 designated herein as psett1 and also confers ethanol resistance to mutant 7e. 1/1
