The present invention relates to a manufacturing method of a transgenic rice callus for mass production of recombinant human acid α-glucosidase (hGAA) with high-mannose glycans for treatment of a Pompe disease and a transgenic rice callus for mass production of human acid α-glucosidase manufactured by the same. According to the present invention, a gnt1 cell line [(ΔGnT I+pMYD85)-01 cell line of rice] (KCTC18586P) for mass production of specific human acid α-glucosidase (hGAA) transformed into a recombinant hGAA expression vector to be manufactured can be provided as a foundation technique of a mass culture system of the glycoprotein hGAA having high-mannose N-glycans to be used in production of a glycoprotein medicine having a mannose terminal by a protein medicine of a Pompe disease which is a lysosomal accumulation disease, and hence can be effectively used in the protein medicine field.
