The present invention relates to a peptide inhibiting the TLR4 signaling pathway, a TLR4 antagonist comprising the peptide, and a pharmaceutical composition, comprising the peptide, for treatment of autoimmune disease or inflammatory disease. The peptide according to the present invention exhibits an excellent effect of suppressing the secretion of inflammatory cytokines and the activation of NF-kB and MAPKs by inhibiting the TLR4 signaling pathway and thus is useful for a pharmaceutical composition for prevention or treatment of autoimmune diseases and inflammatory diseases mediated via the TLR4 signaling pathway.
