Production in Large Animals of the Antibody to the Australia Antigen.

The principal objective reached in the progress of the work performed under the contract has been to demonstrate that it is possible to prepare a heat-treated concentrate of HAA and use this preparation to produce anti-HAA in large animals goats,horses and sheep. Immunization of guinea pigs with HAA positive plasma and heat-treated HAA was successful but no anti-HAA was produced using AB-AG complex as the immunogen. Immunization of hampsters by the ascites fluid method was also not successful. Sheep were found to be the most useful animal since all three immunized produced anti-HAA after I.M. administration of heat-treated HAA, plasmaphoresis of sheep is relatively easy to perform. Several new or modified techniques have been developed which have been useful in accomplishing these studies. The most important of these has been the use of polyethylene glycol to augment antigen-antibody reactions,both in immunoassays and adsorption of antisera prepared in animals.
