Toxicologic Evaluation of Intravenously-Administered Streptozotocin (NSC-85998) in BDF1 Mice

In BDF1 mice administered a single intravenous dose of streptozotocin, estimates of the LD90, LD50, and LD10 could not be reliably ascertained for either male or female mice because of the erratic and irreproducible mortality patterns. Estimates in BDF1 mice administered five daily intravenous doses of streptozotocin could not be determined for the same reason as the single dose study. Toxicity was similar in both sexes in both the single dose and five daily dose schedules. A single dose toxicity study was performed using doses of 350 and 450 mg/kg. Dose dependent toxicity to the gastrointestinal tract, liver, pancreas, thymus, and urogenital system was observed in both sexes as well as in the gonads of males in the single dose study. Histopathologically confirmed pancreatic, hepatic, and urogenital lesions were confirmed by elevations in glucose, BUN, and SGOT. Severe weight loss observed in both sexes was probably related to dehydration from polyuria. Five daily dose toxicity was not performed.
