From rat and human blood concentrates erythrocyte 'ghosts' were prepared, containing membrane-bound acetylcholinesterase (AChE). Solubilized AChE was obtained by treating these ghosts with Triton X-100. After inhibition of AChE with soman, the reactivation by HI-6 (30 micrometers) and aging of the inhibited enzyme in the absence and presence of atropine (0.01 - 1 micrometer) were studied. Soman-inhibited membrane-bound and solubilized AChE from rat and human erythrocytes behaved similarly with regard to reactivation by HI-6 and aging. Atropine did not affect reactivation by HI-6 but using 1 micrometer, it reduced the rate constant of aging of the inhibited enzyme from both preparations.
