Side Effects of Compounds Used in the Treatment of Organophosphate Poisoning

With an open field test, a motor coordination test, a shuttlebox task, quantitative EEG measurements and visual evoked responses, the effects of stepwise increased single doses of atropine, aprophen, P2S, obidoxime and HI-6 were determined. The objective was to obtain an impression above which dose-level these drugs cause significant effects in a subject unchallenged with an organophosphate. Given a conversion factor 7 between rats and men, often used in pharmacological or toxicological Practice, the 'no effect' (NE) doses of atropine, obidoxime and P2S thus determined came close to the sign-free doses established in men. Similar human data on aprophen and HI-6 are not available. It is also clear from these investigations that no single test suffices to find NE and effective (EFF) doses of these compounds. It is concluded that the procedures followed in rats provide a reasonable first estimate of the NE and EFF doses for humans. However, to be on the safe side and for a more precise estimate, confirmation of the results in another species remains necessary. (Copyright (c) 1987 TNO Medical Biological Laboratory.)
