Dimethylamine-2-2ethyl azide (DMAZ, 99.3% pure) was tested for its potentials to induce micronucleated polychromatic erythrocytes (MPCE) in the bone marrow cells of male and female CD-1 mice according to OECD TG 474 in compliance with Good Laboratory Practice. Male and female mice were dosed orally at 125, 250, and 500 mg/kg. Animals were euthanized approximately 24 and 48 hrs after dosing. The percentage of polychromatic erythrocytes (PCE) and the frequency of micronucleated polychromatic erythrocyte (MPCE) were determined at 24 and 48 hrs. Cytotoxicity was assessed by scoring the number of PCE and norchromatic erythrocyte (NCE) in first 200 erythrocytes for each animal. There were no statistically significant increases in the number of MPCE in the treated groups when compared to the concurrent vehicle control group. These results indicate that DMAZ did not cause chromosome damage in the in vivo mouse micronucleus assay and therefore it is not considered to be a clastogenic agent.
