Yields of coenzyme Q10 (CoQ10) after fermentation of Agrobacterium tumefaciens are dependent on the effectiveness of the cell lysis technique used. This study investigated various cell lysis methods, including ultrasound, repetitive freezing/thawing, grinding and acid-heat treatment. Acid-heat treatment using HCl was found to be the most effective means of releasing CoQ10, followed by the same treatment with lactic, sulfuric, phosphoric and oxalic acids. The most significant processing parameters, namely the ratio between acid solution vol. and bacteria wt. (A/B ratio), incubation temp. and reaction time were optimized by central composite design with a quadratic regression model built using response surface methodology. The highest CoQ10 yield (1.518 mg/g dry cell) was achieved using 3 mol/l HCl at an A/B ratio of 10.8 ml/g at 84.2 degrees C for 35.3 min.
