Highlights ? Appling for the method of UPLC–MS/MS to determine the three lignin monomers in wheat straw. ? We have optimized the conditions of chromatography system and MS. ? Realized high-throughput due to improvement of the nitrobenzene oxidation method. Abstract A method to determine the lignin monomers (p-hydroxybenzaldehyde, vanillin and syringaldehyde) in plant cell wall of wheat internode was developed and validated using a high-throughput nitrobenzene oxidation step and ultra performance liquid chromatography tandem mass spectrometry (UPLC–MS/MS) for quantification.
