To further clarify the antitumor mechanism of underlying apoptotic from the molecular level, provide a clinical guidance and lay a foundation for the development of novel, effective antitumor Traditional Chinese Medicine (TCM) agents and pave a path for usage of TCM resources. In this study, the immune effects of MPFP were firstly tested and compared with MP, PP by lymphocyte proliferation. Then, activation experiments of RAW264.7 cell were carried out by detecting proliferation, phagocytosis and secretion of nitric oxide and immune related factors in serum in H22 tumor-bearing mice. To further explore the possible antitumor mechanism of the molecular, protein expression of Bcl-2, caspase-3, and caspase-9 in the thymus and spleen tissues in H22 tumor-bearing mice was determined by Western blot analysis. The MPFP could active RAW264.7 cells in vitro and increase the rate of CD4+ and CD8+ cells in spleen tissues in H22 tumor-bearing mice. The serum high levels of TNF-alpha and IL-2 were observed in mice given with MPFP and Western blot assay showed that MPFP may increase Bcl-2 protein levels and decrease caspase-3 and -9 protein levels in the thymus and spleen tissues, especially for low dose of MPFP. These data collectively indicate that MPFP has both in vivo and in vitro immune effects. In addition, MPFP altered the protein expression of the mitochondria in immune organs, which is another potential pathway for the antitumor activity of enhancement of host's immune system. These findings may provide a basis for the development of fermentation of polysaccharides of anticancer TCM candidates.
