To study on the anti-tumor effect and mechanism of pseudobulbus cremastrae seu pleiones polysaccharide on mice with H22 hepatocellular carcinoma. The solid tumor of H22 were established to observe the anti-tumor effect of pseudobulbus cremastrae seu pleiones polysaccharide. The thymus index and spleen index were used to analyze the influence. The serum interleukin-2 (IL-2) and tumor necrosis factor-alpha (TNF-α) levels were detected by ELISA.SABC Immunohistochemical method to observe the Bcl-2 expression level. The solid tumor inhibition ratio of low, middle and high dose of pseudobulbus cremastrae seu pleiones polysaccharide treated groups were 43.74 %,37.57%,30.76%,respectively, Increases in serum IL2,TNFαactivity, could control the expression of Bcl-2. pseudobulbus cremastrae seu pleiones polysaccharide could inhibit the H22 solid tumor on mice with not a dose-dependent and no obvious side effect. The probable route of inhibitory effect of it on mice with H22 solid tumor is immune-enhancement and acceleration of cell apoptosis.%研究山慈菇多糖对H22肝癌实体瘤小鼠的抗肿瘤作用及其机制。制作小鼠H22实体瘤肝癌模型,通过脾脏指数、胸腺指数分析其对小鼠免疫器官的影响,Elisa检测血清白介素-2(IL-2)含量、肿瘤坏死因子(TNFα)含量,免疫组化法观察(Bcl-2)表达水平。山慈菇多糖的低、中、高剂量组抑瘤率分别为43.74%、37.57%、30.76%,;能增强血清中IL2、TNFα活性;抗凋亡因子Bcl-2的表达量不同程度地减少。山慈菇多糖对H22肝癌小鼠具有一定的肿瘤抑制作用,本次实验不体现剂量依赖性,显示山慈菇多糖无毒副作用,能增强机体的抗肿瘤能力,降低肿瘤的持续增殖能力。
