摘要

Cellulose broken from textile fabric surface using cellulase enzyme result in considerable improve the fabric quality. Wider range sources available for isolation of cellulase producing microorganisms. In this research we focused on isolation cellulase producing microorganism from soil, production of cellulase enzyme by submerged fermentation process and its characterization. The isolated bacteria were then characterized for its morphological, biochemical characters, Molecular characterization using16S rRNA sequencing and BLAST search. The physical and nutritional parameters of fermentation like Concentration of inoculum, Time, pH, temperature, and substrate, were optimized. The produced enzyme further purified using ultra Filtration, precipitationand dialysis was followed. Enzyme activity determined using IUPAC recommended standard protocols. Common quicker and objective methods proposed to optimize the concentration of enzyme, pH, temperature and treatment time. The effectiveness of cellulase treatment evaluated using standard assessment protocols of pilling, bursting strength and weight loss of the fabric.
