Grapes and wine are frequently contaminated with pesticide residues. For the determination of pesticide residues commonly accepted work-up procedures are usually employed. These include solvent extraction and purification of the extracts. Nowadays the direct measurement without work-up procedure is also performed. For a facile analysis LC-MS/MS is an appropriate method to determine many compounds sensitively and with low effort. Therefore direct measurement of 50 pesticides in wine is compared with common work-up procedures to evaluate the suitability. By direct measurement and by the QuEChERS method reliable results are obtained. Both methods are well suited for this purpose. For screening direct measurement would be preferred due to the lower effort and the highest sensitivity. The QuEChERS method should be used for the confirmation of suspicious results. The ChemElut method provided unsatisfactory results, both for recovery studies and for the analysis of real samples.
