Anantigenicsubstancewasisolatedfromratvisceralyolk-sacendodermofthe18th-20thdaysofgestationbyextractionwiththenonionicdetergentNonidetP-40,SephacrylS-300gelfiltration,andRicinuscommunisagglutininaffinitychromatography.Therabbitantiserumdirectedagainstthisantigenicsubstancewheninjectedintopregnantratsduringtheperiodoforganogenesiscausedabnormalembryonicdevelopment,fetalgrowthretardation,andembryonicdeath.Ouchterlonygeldiffusionanalysisdemonstratedthattheantiserumformedoneimmunoprecipitinbandagainstthecrudedetergentextractandacompleteidentitybetweenthepresentvisceralyolk-sacantigenandtherenalglycoproteinantigenpreviouslyisolated(C.C.K.Leung,(1982)J.Exp.Med.,156,372a€“384).Theantigenelutedfromtheantibodyaffinitycolumnappearedtoconsistoftwomajorpeptidesof60and30kDawhenanalyzedbySDS-polyacrylamidegelelectrophoresis.Indirectimmunofluorescentandimmunoperoxidaselocalizationstudiesatthelightmicroscopicleveldemonstratedthatbothratrenalproximaltubuleandembryonicvisceralyolk-sacendodermatvariousgestationalstages(includingtheorganogeneticperiod)sharedthesameantigen.Indirectimmunoperoxidaselocalizationstudiesattheelectronmicroscopicleveldemonstratedthattheantigenwasapartof(orassociatedwith)themicrovillarmembraneandmembraneinvaginationsatthebaseofthemicrovillioftherenalproximaltubuleandvisceralyolk-sacendoderm.Invivoimmunoperoxidaselocalizationstudiesdemonstratedthattheteratogenicantibodieslocalizedwithinthelargephagolysosomesandtheapicalvesiclesofthevisceralyolk-sacendoderm.Itispostulatedthatvisceralyolk-sacpathologywasinducedbytheantibodies.
