Thepresenceofnervegrowthfactorreceptorsandtheimipramine-sensitiveuptakeofcatecholaminesinsympatheticneuronsofchickembryoswereinvestigatedbyautoradiography.Neuronsweredissociatedfromparavertebralgangliaofdifferentembryonicagesandreceptorsandcatecholamineuptakewerethendeterminedbothatthebeginningofcultureandafter2daysinculture,atwhichtimethenumberofsurvivingneuronsisdeterminedbythesurvivalfactorspresent.Itwasfoundthatwhileessentiallyalltheneuronsspecificallybound125I-NGFbothafterdissociationandattheendofthecultureperiod,only60%oftheneuronstakeup[3H]norepinephrineafterdissociation,andthisproportionremainedconstantthroughthecultureperiodunderconditionswherealltheneuronssurvived.AlloftheneuronsmaintainedbyNGFinculture(35%ofthetotal)displayedthisuptake,andincontrast,onlyone-quarterofthosemaintainedbyheartcell-conditionedmediumalone(60%ofthetotal)tookupcatecholamines.TheuptakewasshowntobeneitherinducedbyNGFnorsuppressedbyheartcell-conditionedmedium.Theseresultssupportthehypothesisthatchicksympatheticgangliacontaindiscretesubpopulationsofneuronswhichmaybeselectedinculturebyvirtueoftheirdifferentrequirementsforsurvivalfactors.
