The sortase-mediated labeling method, whose applications have been limited to C-terminal labeling of cell surface proteins, has been expanded for site-specific N-terminal labeling of cell surface proteins.rnCell surface proteins, such as epidermal growth factor receptors (EGFRs) and G protein-coupled receptors (GPCRs), play crucial roles in many cellular processes, and characterization of their functions and spatiotemporal dynamics can lead to identification of the causes of diseases and the development of new drugs. In recent years, many researchers have been developing methods for cell surface protein labeling, which enable site-specific incorporation of chemical probes into target cell surface proteins for fluorescent imaging, and applying these methods to the characterization of cell surface proteins.
