摘要

In this paper we report a perfusion culture microchambernarray chip with a serial dilution microfluidic network fornanalyzing drug dose response over a concentration rangenspanning 6 orders of magnitude, which is required fornpractical drug discovery applications. The microchambernarray chip was equipped with a pressure-driven interface,nin which medium and drug solution were added with anmicropipet and delivered into the microfluidic networknby pneumatic pressure. We demonstrated that the microchambernarray chip could be used to estimate the 50%ngrowth inhibitory concentration using the model anticancerndrug paclitaxel and the model cancer cell line HeLa.nThe results obtained by using the microchamber arraynchip were consistent with those obtained by a conventionalnassay using microplates. The microchamber array chip,nwith its simple interface and well-designed microfluidicnnetwork, has potential as an efficient platform for highthroughputndose response assays in drug discoverynapplications.
