miR-145 miR-155在乳腺癌患者血浆中的表达及临床意义

摘要

目的 探讨miR-145、miR-155在乳腺癌外周血浆中的表达与乳腺癌临床病理特征的关系.方法 2013年10月至2014年2月经病理诊断明确的乳腺癌患者19例为乳腺癌组,健康女性19例为对照组.通过荧光实时定量PCR法检测血浆中miR-145、miR-155的表达水平,分析其与乳腺癌临床病理特征间的相关性.结果 乳腺癌组较对照组miR-155相对表达量升高,差异有统计学意义(P<0.05).两组miR-145相对表达量比较,差异无统计学意义(P>0.05).乳腺癌淋巴结转移阳性与阴性患者miR-145、miR-155相对表达量差异有统计学意义(P<0.05);不同分子分型,雌、孕激素受体阳性与阴性患者miR-155相对表达量比较,差异有统计学意义(P<0.05).结论 血浆microRNA有望成为乳腺癌新的生物标记物,miR-145、miR-155可用来早期预测乳腺癌患者淋巴结转移状态;miR-155可早期预测乳腺癌患者雌、孕激素受体状态及分子分型.%Objective To investigate the expressions of miR-145 and miR-155 in plasma of patients with breast cancer and explore the possibility of miR-145 and miR-155 in plasma as markers of beast cancer. Methods 38 cases(breast cancer 19 cases,healthy control subjects 19 cases)were enrolled from October 2013 to February 2014. The expression levels of miR-145 and miR-155 in plasma were measured by real-time quantitative polymerase chain reaction(real-time qPCR)analysis. The relationships between the expression level of mi-145 and miR-155 in plasma and the feature of clinical pathology were analyzed. Results Compared with the normal control group,plasma expression of miR-155 was significantly up-regulated in breast cancer patients(P<0.05),while no statistically significant difference was found in plasma levels of miR-145(P>0.05).The content of miR-145 and miR-155 between lymph node metastasis and no lymph node metastasis breast cancer,the content of miR-155 among different molecular type,estrogen receptor(ER)status and progesterone receptor(PR)status were significantly different(P<0.05). Conclusion Plasma microRNA is a kind of novel biomarker for breast cancer,miR-145 and miR-155 can be used for the early prediction of lymph node metastasis. MiR-155 can be used for the early prediction of the status of estrogen receptor,progesterone receptor and molecular type in breast cancer patients.
