毛酸浆果醇提物抑菌机理的初步研究

     

摘要

To investigate the in vitro antibacterial mechanism of the alcohol extract of the fruit of Physalis pubescens L (AEFPP), the growth curve of the Escherichia coli and the changes of the ultrastructure were observed. Staphylococcus aureus and Escherichia coli strains as the indicator bacteria standards were studied by drawing the growth curve and observe the changes in different time points and show ultrastructure of bacteria change by SEM. Under the action of minimum inhibitory concentration (MIC) of the alcohol extract of the fruit of Physalis pubescens L OD value in the control group the figure was below 0.5 throughout. In TEM, in Oh, mitotic figures of the bacteria could be seen obviously; 4h later, vacuoles emerged and the mitotic figures were decreasing; 8h later, a collection of bacteria was killed. In vitro studies have demonstrated that AEFPP inhibits the growth of Escherichia coli which emerges plasmolysis, cytoplasmic vacuolation and cellular death.%通过观察大肠埃希菌标准菌株、金黄色葡萄球菌标准菌株生长曲线和菌体超微结构的变化,探讨毛酸浆果醇提物的可能抑菌机理.以金黄色葡萄球菌标准菌株和大肠杆菌标准菌株作为指示菌,在分别绘制细菌生长曲线的基础上,于不同时间点观察加药后细菌生长曲线的变化,并以透射电镜观察细菌菌体超微结构的改变.在毛酸浆果醇提物最低抑菌浓度作用下,大肠埃希菌标准菌株生长曲线的各时间点菌体浓度OD值始终保持在0.5以下.透射电镜下可见:大肠埃希菌标准菌株生长0h时,细菌出现明显的分裂相;4h后,大肠埃希菌菌体内部出现许多空泡,分裂相明显减少;8h后,出现较多死亡菌体.毛酸浆果醇提物能有效地抑制大肠埃希菌的生长,使大肠埃希菌的菌体结构出现质壁分离、空泡样变和死亡现象.
