高效液相色谱-四级杆飞行时间串联质谱分析3种荷花的花色苷

         

摘要

应用高效液相色谱-电喷雾/四极杆飞行时间串联质谱联用技术(HPLC—ESI/Q—TOF—MS/MS)分析了3种荷花(大红袍、洪湖红莲、红牡丹)的花色苷成分。首先以φHCl=0.1%的甲醇超声提取荷花中的花色苷,然后用XAD-7大孔树脂纯化,选用AmethystC18色谱柱(250mm×4.6mm,5μm),以甲醇-水(φHClHF3I3=0.05%)为流动相进行梯度洗脱,柱后流出液采用电喷雾四极杆飞行时间质谱(ESI/Q—TOF-MS/MS)进行检测,共分离鉴定出6种花色苷,并且运用pH示差法分别测定了3种荷花中总花色苷的含量,为开发利用这一丰富的自然资源提供一定的参考依据。%We employed HPLC-Electrospray Ionization/Quadrupole-Time of Flight-Mass/Mass Spectrometry ( HPLC-ESI/ Q-TOF-MS/MS) to analyze the anthocyanins in three species Nelumbo nucifera. The anthocyanins were extracted with methanol containing HCI (φFacl = 0. 1%) and then purified on an Amberlite XAD-7 column. We applied an Amethyst C18 column (250 mm ×4.6 mm, 5 μm) to HPLC analysis with water methanol-water (containing trifluoroacetic acid (φC1HF3O3 = 0.0.5% ) ) in gradient as mobile phase. The eluates were then detected by ESI/Q-TOF-MS/MS. We altogether identified six s.pecies anthocyanins from these Nelumbo nucifera. We further determined their contents by pH-differential spectrophotometry. It provides theoretical foundation for further development and utilization of Nelumbo nucifera resources.
