茵陈蒿汤防治大鼠非酒精性脂肪性肝病效果及其对肝组织NTCP表达的影响

         

摘要

Objective To evaluate the efficacy of Yinchenhao decoction in prevention and treatment of non-alcoholic fatty liver disease ( NAFLD) in rats, and to study its regulation on sodium taurocholate cotransporting polypeptide ( NTCP) expression and explore its mechanism. Methods Forty-five SD rats were randomly divided into three groups after one week adaptive feeding: the control group ( n = 15) , the high-fat diet model group ( n = 15) , and the Yinchenhao Decoction intervention group ( n = 15) . All rats were sacrificed in batches after 12 weeks, and the levels of blood sugar, body weight, liver weight, hepatosomatic index, as well as serology examinations of glutamic pyruvic transaminase, glutamic oxaloacetic transaminase, triglyceride and bile acid were detected. The pathological changes were evaluated by Hematoxylin and Eosin ( HE) staining; the reverse transcription polymerase chain reaction ( RT-PCR) and Western blotting were used to detect the expression of NTCP gene. Results Compared with the control group, the levels of ALT, AST, TBA, and TG all significantly increased in the model group ( all P 0. 05) . HE staining showed that vacuolar degeneration, inflammatory cell infiltration and brown-yellow bile droplets were found in the liver tissues in the model group.Whereas, these pathological changes all relieved to some degree in the intervention group. The mRNA and protein expression of NTCP was significantly down-regulated in the model group as compared with that of the control group ( P 0. 05) . Conclusion Yinchenhao decoction may improve the liver function and relieve pathological damage by up-regulating the expression of NTCP, and thus alleviate the cholestasis of NAFLD rats.%目的 观察茵陈蒿汤防治大鼠非酒精性脂肪性肝病 (NAFLD) 的效果及其对肝组织钠离子牛磺胆酸共转运多肽 (NTCP) 表达的影响, 探讨茵陈蒿汤防治NAFLD的机制.方法 将45只SD大鼠随机分为3组各15只, 模型组、干预组每天给予高脂饲料灌胃进行NAFLD造模, 同时干预组每天给予茵陈蒿汤10 m L/kg灌胃, 对照组每天给予普通饲料喂养.12周后分批处死, 取大鼠腹主动脉血及肝脏组织;生化分析仪检测丙氨酸转氨酶 (ALT) 、天门冬氨酸转氨酶 (AST) 、总胆汁酸 (TBA) 、甘油三酯 (TG) , HE染色观察肝组织病理变化; Real time PCR、Western blotting法分别检测肝脏组织中NTCP mRNA及蛋白表达.结果 与对照组比较, 模型组血清ALT、AST、TBA、TG水平均升高 (P均 0. 05) .HE染色显示, 模型组肝组织可见空泡样变性、炎性细胞浸润和棕黄色胆汁颗粒, 而干预组较模型组肝组织脂肪变性、炎性细胞浸润均有一定程度缓解.与对照组比较, 模型组肝组织中NTCP mRNA及蛋白表达水平均下调 (P均 0. 05) .结论 茵陈蒿汤可能通过上调NTCP基因表达, 减轻肝细胞内胆汁淤积, 进而改善肝脏功能, 减轻NAFLD大鼠肝脏病理损害.
