大肠癌组织中TAMs、TGF-β1表达与上皮间质转化的关系

             

摘要

Objective To investigate the relationship between tumor-associated macrophages(TAMs) , transforming growth factor β1 (TGF-β1 ) expression and epithelial mesenchymal transition ( EMT) in colorectal cancer tissue. Method Immunohistochemistry was used to detected the expression of TAMs marked CD68 , TGF-β1, E-cadherin and Vimentin in 80 colorectal cancer specimens and matched adjacent non-cancerous tissue specimens. Result In colorectal cancer tissue, the TAMs count was 30.6 ±8.2 /HP, E-cadherin expression was positive in 31cases(38. 8% ), TGF-pl expression was positive in 58 cases(72.5% ), Vimentin expression was positive in 38 cases(47. 5% ). Bui 16.4 ±7. 2 /HP, 64 cases (80.0% ) ,23 cases(28.8% ), 0 in adjacent non-cancerous tissue. TAMs count, TGF-β1, E-cadherin and Vimentin protein expression were correlated with differentiation, depth of invasion, Duke stages and lymph node metastasis in colorectal cancer, all P<0.05. TAMs count, TGF-pl expression was negatively correlated with E-cadherin protein expression (r = -0. 64 and - 0. 39, P < 0. 01 ) while positive correlated with Vimentin protein expression ( r = 0. 62 and 0. 33, P < ? 0.05). Conclusion The infiltration of TAMs and TGF-pl overexpressed caused inflammatory environment maybe involved in EMT in colorectal cancer.%目的 探讨大肠癌组织中肿瘤相关巨噬细胞(TAMs)、转化生长因子β1(TGF-β1)表达与上皮间质转化(EMT)的关系.方法 采用免疫组化法检测80例大肠癌及癌旁正常组织中CD68标记的TAMs、TGF-β1、上皮标志物E-cadherin、间质标志物Vimentin蛋白.结果 大肠癌组织中TAM计数为(30.6±8.2)个/HP,TGF-β1蛋白阳性58例(72.5%),E-cadherin蛋白阳性31例(38.8%),Vimentin蛋白阳性38例(47.5%);而癌旁正常组织中分别为(16.4±7.2)个/HP、23例(28.8%)、64例(80.0%)、0例.两者比较,P均<0.01.TAMs计数及TGF-β1、E-cadherin、Vimentin蛋白表达与大肠癌分化程度、浸润深度、Duke分期和淋巴结转移有关(P均<0.05).TAMs计数、TGF-β1与E-cadherin蛋白表达呈负相关(r值分别为-0.64、-0.39,P均<0.01),而与Vimentin蛋白表达成正相关(r值分别为0.62、0.33,P均<0.05).结论 大肠癌组织中TAMs浸润及TGF-β1高表达所造成的炎症环境可能与大肠癌的EMT有关.
