β-榄香烯对肾癌786-0细胞凋亡的影响及其机制探讨

Objective To investigate the effect of β-elemene on apoptesis in human renal carcinoma cell 786-0 and the mechanism involved. Methods 786-0 cells were treated with different concentrations of β-elemene for 24 h, cell viability was measured by MTT assay, and apoptosis was evaluated by flow cytometry. Western blot was employed to detect the expression of protein kinase B (p-Akt) and total Akt. Results β-elemene inhibited the viability of 786-0 cells in a dosedependent manner; it also induced apoptosis and inhibited the expression of p-Akt ( all P ＜ 0. 05 ). Conclusion β-elernene can induce apoptosis in human renal carcinoma cell 786-0, inhibiting PI3K/Akt signaling pathway maybe the mechanism.%目的 观察β-榄香烯对肾癌786-0细胞凋亡的影响,并探讨其可能机制.方法 使用不同浓度β-榄香烯作用于786-0细胞24 h,MTT法检测细胞活力,流式细胞术检测细胞凋亡率,Western blot法检测细胞内磷酸化蛋白激酶B(p-Akt)及总Akt.结果 随β-榄香烯浓度增大,细胞存活率逐渐降低,细胞凋亡率明显增加,细胞内p-Akt表达水平逐渐降低,均呈一定浓度依赖性(P均<0.05).结论 β-榄香烯可诱导肾癌细胞凋亡,其机制可能为抑制PI3K/Akt通路活性.
