HIF-2α在宫颈癌组织中的表达及意义

摘要

目的：探讨低氧诱导因子2α（HIF‐2α）在宫颈癌组织中的表达以及在宫颈癌发生发展中的作用。方法：采用免疫组化法检测 HIF‐2α在正常宫颈及宫颈癌组织中的表达，并将 HIF‐2α表达与宫颈癌的临床病理特征进行相关分析；采用siRNA技术干扰HIF‐2α在宫颈癌细胞 HeLa、SiHa的表达，Western blot技术检测HIF‐2α蛋白在宫颈癌细胞的表达；M T T试验检测HeLa、Si‐Ha细胞增殖情况。结果：HIF‐2α在宫颈癌组织的表达明显高于正常宫颈组织，经χ2检验，有统计学意义，并且HIF‐2α表达与宫颈癌的淋巴转移呈正相关，但与宫颈癌患者的年龄、病理分级以及临床分期无显著性相关。低氧可诱导 HIF‐2α表达增加，并促进 HeLa、SiHa细胞增殖，而干扰 HIF‐2α表达后，则抑制HeLa、SiHa细胞增殖，经 t检验，具有统计学意义。结论：HIF‐2α在宫颈癌中的表达升高，其对宫颈癌的发生发展起促进作用。%Objective:To explore the expression of HIF‐2α in cervical cancer and its role in the cervical cancer initiation and progression .Methods:The expression of HIF‐2α in cervical cancer and normal cervix were measured using immunohistochemistry ,and the correlation between HIF‐2αand clinicopathological parameter of cer‐vical cancer was analyzed .HIF‐2αexpression in HeLa and SiHa cells were measured by western blot assay and HIF‐2αin HeLa and SiHa cells were knocked down by siRNA technology .Subsequently ,cell proliferation assay were performed by MTT .Results:The expression of HIF‐2αin cervical cancer was significantly increased compared with the normal cervix ,and HIF‐2αexpression was positively correlated with lymphatic metastasis of cervical cancer ,but the significant correlation between HIF‐2αexpression and age ,pathological grade ,clinical stage of patients were not observed .Hypoxia induced the expression of HIF‐2αand promoted cell proliferation of HeLa and SiHa cells ,but the knockdown of HIF‐2αresulted in a significant inhibition of cell proliferation .Conclusion:HIF‐2αexpression in cervi‐cal cancer is significant increased ,and HIF‐2αpromoted the initiation and progression of cervical caner .
