微小RNA多态性和非小细胞肺癌遗传易感性关系研究

             

摘要

Objective:To study the relationship between micro RNA polymorphism and non small cell lung cancer genetic susceptibility.Methods:130 patients (group A) with non small cell lung cancer and 150 healthy individuals (group B) were selected Rs2910164 hsa-mir-146a and Rs3746444 hsa-mir-499 were selected to study the polymorphism of the gene, and the OR and 95%CI were calculated by using the non conditional ogistic regression model.Results:The gene ratio of G and A in group A were 73.08% and 26.92% respectively, while in group B were 77.33% and 22.67%;the genotype frequencies of GG, AG and AA in group A were 52.31%, 40.77%, 6.92%, and in group B were 58.67%, 36.67%, 4.66%, the genotype and allele distribution between two groups were no significant differences(P=0.4892, P=0.2437);adjusting for sex, age, smoking, drinking and regression analysis, in hsa-mir-146a rs2910164, genotype CC patients with higher risk of non-small cell lung cancer compared to the genotype GG+CG patients (P=0.0126);in hsa-mir-499 rs3746444,;after correction of various factors, with CC genotype male Patients with higher risk of non small cell lung cancer (P=0.0369) and smoking patients with higher risk of non small cell lung cancer (P=0.0419), drinking for higher risk of non small cell lung cancer (P=0.0364).Conclusion:The gene type CC in Micro RNA rs2910164 may increase the genetic susceptibility to non small cell lung cancer, and the risk of the non small cell lung cancer disease and the factors of smoking and drinking have a certain relationship.%目的:研究微小RNA多态性和非小细胞肺癌遗传易感性的关系.方法:选取非小细胞肺癌患者130例(A组)与常规体检者150例(B组),对两组hsa-mir-146a rs2910164和hsa-mir-499 Rs3746444基因的多态性进行研究分析,利用非条件Logistic回归模型计算OR及95%CI等.结果:A组G、A基因比例分别为73.08%和26.92%,B组分别为77.33%和22.67%;A组GG、AG、AA基因型频率分别为52.31%、40.77%、6.92%,B组分别为58.67%、36.67%、4.66%;两组基因型与等位基因分布分别比较均无统计学差异(P=0.4892, P=0.2437);调节性别、年龄、吸烟、饮酒等进行回归分析,在hsa-mir-146a rs2910164中,基因型CC与基因型GG+CG相比患非小细胞肺癌的危险性较高(OR=1.39, 95%CI:1.06~1.80, P=0.0126);在hsa-mir-499 Rs3746444中,两组基因型频率分布比较无统计学差异(P>0.05);校正各种因素后,带有CC基因型男性患非小细胞肺癌危险性较高(OR=1.61, 95%CI:1.24~2.03, P=0.0369),吸烟患非小细胞肺癌危险性较高(OR=2.01, 95%CI:1.76~2.35, P=0.0419),饮酒患非小细胞肺癌危险性较高(OR=1.22, 95%CI:0.55~3.25, P=0.0364).结论:微小RNA rs2910164中基因型CC有可能增加非小细胞肺癌的遗传易感性,且其发病风险与吸烟、饮酒等因素存在一定相关性.
