促肾上腺皮质激素对创伤性休克大鼠急性肺损伤的保护作用及其机制

         

摘要

目的:探讨促肾上腺皮质激素(ACTH)对创伤性休克(TS)大鼠急性肺损伤的影响及其相关机制。方法 Wistar大鼠36只,随机分成假手术组(C组)、模型(TS)组和ACTH组,每组12只。检测血清肿瘤坏死因子(TNF)-α和肺组织超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)的含量;计算肺湿干重比(W/D);光镜下观察肺组织病理变化,并做病理评分。结果与C组比较,TS组与ACTH组肺组织匀浆MDA及血清TNF-α水平、肺组织W/D及损伤评分均明显升高(P<0.01);肺组织匀浆SOD及GSH活性均降低(P<0.01)。与TS组比较,ACTH组血清TNF-α及肺组织匀浆MDA水平、肺组织W/D及损伤评分均明显降低(P<0.01或P<0.05),SOD及GSH活性均升高(P<0.01)。结论 ACTH预处理可减轻TS大鼠急性肺损伤,可能与其抑制氧自由基产生及促炎细胞因子的释放有关。%Objective To investigate the effects of andrenocorticotrophic hormone (ACTH) on acute lung injury in rats with traumatic shock (TS) and explore the underlying mechanism. Methods The 36 male Wistar rats were assigned randomly into 3 groups(n=12,in each): sham operation group(C),TS model group and ACTH pretreatment group. The arterial blood samples were taken for determination of serum levels of tumor necrosis factor-α (TNF-α). The lung tissues were removed for determination of levels of superoxide dismutase (SOD), malondialdehyde (MDA) and glutathione (GSH) and microscopic examination. Results Compared with group C, the serum levels of TNF-α, the lung levels of MDA,W/D lung weight ratio and pathologic scores of lung tissue were significantly higher (P<0.01),and the lung levels of SOD and GSH in group TS and group ACTH were significantly lower (P<0.01). The serum level of TNF-α, the lung levels of MDA,W/D lung weight ratio and pathologic scores of lung tissue were significantly lower (P<0.01 or P<0.05),and the levels of SOD and GSH were significantly higher (P<0.01) in group ACTH than those in group TS. Conclusion The supplementation of ACTH can ameliorate acute lung injury in rats with TS, which may contribute to the inhibition of oxygen free radicals and inflammatory cytokines expression.
