咖啡酸苯乙酯对白化病大鼠视网膜光损伤的保护研究

         

摘要

目的 从形态和功能两方面测试咖啡酸苯乙酯(CAPE)是否能够保护视网膜组织免于光损伤诱导的氧化损伤.方法 分别将大鼠暴露于50 lx循环暗光2周、50 lx循环暗光8周、200 lx循环明光8周,同时给予0.01% CAPE和对照饲料喂养;以ERG和组织学分别检测大鼠视网膜对循环暗光和循环明光的反应.结果 不论是喂养2周还是喂养8周,在循环暗光下,CAPE组ERG A/B波幅均显著高于对照组(P<0.05);在循环明光下,ERGA/B波幅CAPE组和对照组之间差异无统计学意义(P>0.05);上述三组组织学结果差异均无统计学意义.结论 PE可以在循环暗光下保护视网膜功能.CAPE有可能作为一种天然化合物保护白化病或者其他视网膜变性患者视网膜功能.%Objective The purpose of this study was to test whether caffeic acid phenethyl ester (CAPE) could protect the retina against chronic dim or bright light damage. Methods Albino rats, raised under dim (50 lux) cyclic light for 2 weeks, dim (50 lux) cyclic light for 8 weeks or bright (200 lux) cyclic light for 8 weeks, were fed with either AIN-96A diet (control) or AIN-96A diet supplemented with 0. 01% CAPE for 2 ~8 weeks. Retinal structure and function were determined by histology and electroretinography ( ERG). Results With chronic dim light induced retinal degeneration system, the ERG of the CAPE-fed retina was significantly higher than that of the control-diet-fed retina in dim light either for 2 weeks or 8 weeks (P < 0. 05). With chronic bright light induced retinal degeneration system, there was no significant difference between the CAPE-fed retina and the control-diet-fed retina (P > 0.05). There were no significant differences in histological results between any two groups. Conclusions CAPE can preserve retinal function in chronic dim light It can be a potential candidate for supplementation in diet for preservation of retinal function from day to day oxida-tive light stress.
