化瘀生肌方提取物对糖尿病溃疡模型大鼠创面愈合的影响

         

摘要

Objective To investigate the effect of Huayushengji extract on wound healing in the diabetic ulcer model of rats and the mechanism.Methods Thirty-two clean SD rats were selected.Twentyfour of them were randomly selected to establish a diabetic ulcer model before they were randomly divided into three groups (n =8):the model group (treated with blank gel), Beifuxingroup (treated with recombinant bovine basic fibroblast growth factor gel after modeling), and TCM group (treated with Huayushengjirecipeextract gel).The remaining eight SD rats served as the control group.Rats in each group were treated for two weeks.Serum levels of basic fibroblast growth factor (bFGF) and epidermal growth factor (EGF) were detected in each group.Morphological changes were observed with HE staining, while the the numbers of capillaries and fibroblasts in granulation tissues were detected by immunohistochemistry.Results The ulcer surface of the TCM group healed the fastest, followed by the Beifuxin group compaired with the model group.The serum levels of EGF and bFGF in the model group were significantly lower than those in the control group, and the difference was statistically significant (P<0.01).The serum levels of EGF and bFGF in the Beifuxin group and TCM group were significantly higher than those in the model group, and the difference was statistically significant (P<0.05, P<0.001).Fourteen days after modeling, the number of capillaries and fibroblasts in the model group significantly decreased, and the difference was statistically significant (P<0.01).After treatment, the number of capillaries and fibroblasts in the Beifuxingroup and TCM group were significantly higher than that in the model group, and the difference was statistically significant (P<0.05, P<0.001).Conclusion Huayushengji recipe can significantly increase the serum levels of EGF and bFGF in rats, which can contribute to the proliferation of new capillaries and fibroblasts, and promote granulation tissue growth.%目的 探讨化瘀生肌方提取物对糖尿病溃疡模型大鼠创面愈合的影响, 并探讨其可能的作用机制.方法32只清洁级SD大鼠, 随机抽24只制造糖尿病溃疡大鼠模型, 造模成功后随机分为3组:模型组 (给予空白凝胶) 、贝复新组 (给予重组牛碱性成纤维细胞生长因子凝胶) 、中药组 (给予化瘀生肌方提取物凝胶), 每组8只.未造模的8只大鼠为对照组.各组大鼠给药2周后, 分别检测血清碱性成纤维细胞生长因子 (bFGF) 、表皮生长因子 (EGF) 水平.HE染色观察各组形态学变化, 免疫组化法检测创面肉芽组织中新生毛细血管数及成纤维细胞数.结果 与模型组比较, 中药组溃疡面愈合较快, 贝复新组次之.模型组大鼠血清EGF、bFGF水平比对照组明显降低, 差异有统计学意义 (P<0.01);贝复新组、中药组大鼠血清EGF、bFGF水平均明显高于模型组, 差异有统计学意义 (P<0.05, P<0.001);造模14d后, 模型组新生毛细血管数密度及成纤维细胞数量明显降低, 差异有统计学意义 (P<0.01);贝复新组、中药组治疗后新生毛细血管数密度及成纤维细胞数均明显高于模型组, 差异有统计学意义 (P<0.05, P<0.001).结论 化瘀生肌方提取物能够使大鼠血清中EGF及bFGF水平明显升高;并利于新生毛细血管及成纤维细胞的增殖, 促进创面肉芽组织生长.
