RP-HPLC法测定藤黄果原料中羟基柠檬酸的含量

         

摘要

Objective To establish an HPLC method for the determination of hydroxycitric acid in Garciniacambogia .Methods The analysis was performed on a Agilent TC‐C18 (250 mm × 4 .6 mm ,5 μm) column with 0 .05 mol · L -1 sodium sulfate solution (ad‐justed pH to 2 .3 with sulfuric acid) as the mobile phase .The flow rate was 1 .0 mL · min-1 ,the detection wavelength was 210 nm ,the injection volume was 10μL ,and the column temperature was 30 ℃ .Results The calibration curve was linear within the range of 0 .209 9‐2 .099 0μg (r=0 .999 9 ,n=6) ,and the average recovery was 99 .2% ,RSD 0 .5% .Conclusion The method was simple ,accurate ,and with good reproducibility .It could be used for the determination of hydroxycitric acid in Garcinia cambogia .%目的:建立藤黄果原料中羟基柠檬酸的 HPLC 测定方法。方法色谱柱Agilent TC‐C18(250 mm ×4.6 mm ,5μm);流动相为0.05 mol · L -1的无水硫酸钠水溶液(用硫酸调节pH为2.3);检测波长210 nm ;进样量10μL ;体积流量1.0 mL · min-1;柱温30℃。结果羟基柠檬酸的线性范围为0.2099~2.0990μg ,r=0.9999(n=6),平均回收率99.2%,RSD为0.5%。结论该方法简单、准确、重复性好,可用于藤黄果原料中羟基柠檬酸的定量测定。
