当归多糖对辐射损伤小鼠血液中microRNA作用的研究

         

摘要

Objective To ingestigate the effects of Angelica polysaccharides on the differential expression profiles of micro RNA in-duced by 60Coγ-ray radiation damage in mice peripheral blood and to explore their significances.Methods 35 SPFC57BL/6Jmice were randomly divived into 7 groups:control group,2Gy no intervention radiation group,2Gy APS 4 mg·kg-1 radiation group, 2 Gy APS8 mg·kg-1 radiation group,10 Gy no intervention radiationg roup,10 Gy APS4 mg·kg-1 radiation group and 10 Gy APS8 mg·kg-1 radiation group.Radiation group mice were exposed to 60Coγ-ray,at the dosage of 2 Gy,10 Gy(dosage rate 10 Gy·h-1)single whole body radiation.After radiation,radiation group mice were injured with APS 4 mg·kg-1 and APS8 mg·kg-1 for 3 days consecutively by injection.Peripheral blood of mice were collected after 72h.The screening of microRNA different ex-pression in peripheral blood was used by Agilent microRNA microarrays.Results Compared with no intervention radiation group, there was totally 30 and 31 microRNA showing differential expression in 2 Gy and 10 Gy no intervention group,respectively.The expression of microRNA was significant difference between control group and no intervention radiation group(P0.05).10 Gy APS 4 mg·kg-1 radiation group and 10 Gy APS 8 mg·kg-1 radiation group increased the range of microRNA expression.Compared with 10 Gy APS 4 mg·kg-1 radiation group,the differential expression levels of microRNA decreased obviously in 10 Gy APS 8 mg·kg-1 radiation group,excepte for microRNA-455,microRNA-9 and let-7,there was statistical significance between 10 Gy APS 4 mg·kg-1 radiation group and 10 Gy APS8 mg·kg-1 radiation group(P0.05).10Gy照射APS4mg·kg-1组和10Gy照射APS8mg·kg-1组治疗差异表达microRNA均有所降低,除microRNA-455、microRNA-9和let-7外,10Gy照射APS8mg·kg-1组治疗降低更加显著,与10Gy照射APS4mg·kg-1组比较差异有统计学意义(P<0.05).结论 APS通过改变血液中microRNA的表达,促进辐射小鼠的损伤修复.
