本文对淡豆豉中异黄酮酸水解产物的乙酰胆碱酯酶抑制作用进行了研究.采用Ellman比色法测抑制率,UPLC测大豆苷、黄豆黄苷、染料木苷、黄豆苷元、黄豆黄素、染料木素的含量,相关性分析筛选其药效成分.异黄酮酸水解的工艺为温度70℃,3 moL/L盐酸甲醇溶液,水解5h,对乙酰胆碱酯酶的抑制率最高,可达到82.37%.相关性分析表明,淡豆豉异黄酮对乙酰胆碱酯酶的抑制率与三种苷元的浓度呈正相关的关系,并且与总苷元的相关性最大达到0.869(P <0.01).淡豆豉中起乙酰胆碱酯酶抑制作用的成分可能为总异黄酮苷元部分.%To study the inhibitory ability of acid-hydrolysate of isoflavones in Fermented Soybean.The ability of acetylcholinesterase inhibitory was determined with Ellman colorimetry method.An UPLC method was established for the simultaneous determination of daidzin,glycitin,genistin,daidzein,glycitein and genistein in fermented soybean.The optimal conditions for the hydrolysis of isoflavones were found to be at 70 ℃ with 3 mol/L HCl methanol for 5 h,and the acetyl cholinesterase inhibition rate reached 82.37%.The acetyl cholinesterase inhibition rate had significant positive correlation with isoflavone aglycone (P < 0.01).
