不同促效剂对唾液腺上皮细胞pIgR表达的影响

         

摘要

Objective It is to investigate the effects of different agonists on the expression of polyimmunoglobulin receptor (pIgR) in salivary gland epithelial cells, and to observe the changes of secretory immunoglobulin Ig A (sIgA), SC, IgA, CD3 cells and CD8 cells. Methods Salivary gland epithelial cell suspensions were randomly divided into control group, adrenaline group, isoproterenol group and carbachol group which were treated with respective medicine for 24 h. The expression of pIgR mRNA in salivary gland epithelial cells was determined by fluorescence quantitative RT-PCR.The changes of sIgA, SC and IgA in each group were measured by ELISA, and the percentages of CD3 and CD8 cells were determined by flow cytometry. Results Compared with the control group, the adrenaline group, isoproterenol group and carbachol group could significantly promote the expression of pIgR mRNA in salivary gland epithelial cells (P<0.05), and the promotion in adrenalin group was more significant (P<0.05).The expression of pIgR, sIgA, SC, IgA and the percentages of CD3 and CD8 cells in each treatment group were significantly higher than those in the control group (P<0.05), the expression of pIgR, sIgA, SC, IgA and the percentages of CD3 and CD8 cells in the adrenaline group was the highest. Conclusion The agonists adrenaline, isoproterenol and carbachol can increase the expression of pIgR in salivary gland epithelial cells, promote the secretion of sIgA, SC and IgA, increase the percentage of CD3 cells and CD8 cells, thus to regulate the local immunity of salivary glands, the effect of adrenaline is better than isoproterenol and carbachol.%目的 探讨不同促效剂对唾液腺上皮细胞多聚免疫球蛋白受体(pIgR)表达的影响,并观察其分泌型免疫球蛋白IgA(sIgA)、分泌游离分泌片(SC)、IgA及CD3+和CD8+水平变化.方法 将唾液腺上皮细胞悬液随机分为空白对照组、肾上腺素组、异丙肾上腺素组、卡巴胆碱组,分别给空置和相应药物处理24 h,采用荧光定量RT-PCR测定各组唾液腺上皮细胞pIgR mRNA水平,采用酶联免疫吸附(ELISA)法测定各组sIgA、SC、IgA水平,流式细胞术测定其CD3+、CD8+细胞百分率.结果 与空白对照组比较,肾上腺素组、异丙肾上腺素组、卡巴胆碱组均能够明显促进唾液腺上皮细胞pIgR mRNA的表达(P均<0.05),其中肾上腺素组对pIgR mRNA表达的促进作用更显著(P<0.05).各给药组sIgA、SC、IgA分泌水平与CD3+、CD8+细胞百分率均明显高于空白对照组(P均<0.05),其中肾上腺素组sIgA、SC、IgA分泌水平与CD3+、CD8+细胞百分率最高(P均<0.05).结论 肾上腺素、异丙肾上腺素、卡巴胆碱等促效剂均可以增加唾液腺上皮细胞pIgR的表达,促进sIgA、SC、IgA分泌,增加CD3+细胞和CD8+细胞百分率,进而达到调节唾液腺局部免疫功能的目的,其中肾上腺素效果优于异丙肾上腺素、卡巴胆碱.
