防风对大鼠和小鼠胃肠运动的抑制作用及机制研究

         

摘要

目的 观察防风(RS)对大小鼠胃肠运动的抑制作用并探讨其机制.方法 采用苯酚红测定法观察RS对小鼠胃肠运动的影响;制备大鼠结肠平滑肌肌条并放置在浴槽中,滴加RS及其他试剂并记录肌条的收缩曲线加以分析;分离并培养大鼠结肠平滑肌细胞,探针孵育后,不同缓冲液条件下测定NS及RS处理前后细胞的荧光值并加以分析.结果 RS各组对小鼠小肠推进和胃排空均有抑制作用;RS各组均抑制离体大鼠结肠平滑肌肌条收缩,抑制Ach引起的肌条收缩.RS剂量与峰高和同线下面积有量效关系.不同缓冲液对RS处理后的荧光值和抑制率有显著性差异.结论 防风可抑制小鼠小肠推进及胃排空,抑制离体大鼠结肠平滑肌收缩,机制与肾上腺素能α受体及M胆碱受体有关,与阿片受体和β受体无关;防风可降低细胞内游离钙离子浓度,其机制与抑制细胞外钙离子内流有关,与抑制细胞内钙离子释放无关.%Objective It is to observe the inhibition of Radix Saposhnikoviae ( RS ) on the small inlestinal and gastric of mice and rat, and approach the possible mechanism. Methods The influence of RS on small intestinal propulsion ( SIP ) and gastric emptying ( GE ) in mice was observed by determination of phenol red. The colonic longitudinal muscle strips of rat were prepared and suspended in organ bath , then RS and the other reagent were dropwised and the contraction of strips was recorded to analysis. The smooth muscle cells from colon of rat were isolated and cultured and loaded with Fluo - 4/AM, and then the fluorescent intensity ( FI ) of intracellular Ca2+ of those cells was measured to analysis before and after treated with RS or NS.Results RS had inhibition on the SIP and GE in mice. RS had inhibition on the contraction of smooth muscle strips and had inhibition against Ach-induced contraction. And it had obvious dose-effect relationship between RS and peak height or AUC.There were significant differences in FI and inhibition after treated with RS between different buffer solutions. Conclusion RS can inhibit GE and SIP in mice. and inhibit spontaneous contraction of colonic smooth muscle strips of rat in vitro. Its possible inhibitory mechanism had close relations with the α - adrenoceptor and perhaps with the M - cholinergic receptor, but had little relation with the β - adrenoceptor and the opioid receptor. RS could prevent the mobilization of Ca2+ from extracellular into intracellular in some degree, but had no effect on the release of Ca2+ from intracellular.
