大肠癌中E2F-1、β-catenin和cyclinD1的表达关系

         

摘要

目的 探讨大肠癌组织中β-catenin异常表达的意义及其与 E2F-1、cyclinD1表达的关系.方法 采用免疫组化SP法检测78例大肠癌、20例正常大肠黏膜、30例大肠腺瘤中E2F-1、β-catenin 和cyclinD1的表达情况.结果 20例正常大肠组织中β-catenin蛋白呈正常表达,而E2F-1、cyclinD1表达呈阴性.大肠腺瘤和大肠癌组织中E2F-1蛋白阳性表达为6.67%和61.54%;β-catenin蛋白异常表达为36.67%和92.31%; cyclinD1蛋白阳性表达为13.33%和55.13%.E2F-1、cyclinD1蛋白阳性表达和β-catenin蛋白异常表达均与大肠癌病理分级、临床病理分期以及淋巴结转移有关(P均<0.05).大肠癌中β-catenin与cyclinD1呈正相关(P<0.05);β-catenin与E2F-1没有明显相关性(P>0.05),但E2F-1和cyclinD1呈明显相关性(P<0.05).结论 E2F-1、β-catenin和cyclinD1的表达与大肠癌的发生、发展及浸润转移有关,可作为判断大肠癌生物学行为和预后的重要指标.%Objective It is to approach the significance of abnormal expression of p - catenin in colorectal cancer tissue, and the relationship between the expression of cyclinDl , E2F - 1 and p - catenin. Methods The immunohistochemical SP method were used to detect the expressions of E2F - 1 , p - catenin and cyclinDl in 78 cases of colorectal adenocarcinoma, 20 cases of normal colorectal mucosa and 30 cases of colorectal adenoma. Results In the 20 cases of normal colorectal tissues, the expression of p - catenin protein was normal, while the expression of E2F - 1 and cyclinDl was negative. In colorectal adenoma and colorectal cancer the positive expression of E2F - 1 protein was 6. 67% and 61. 54% , the abnormal expression of p - catenin protein was 36. 67% and 92.31%, the expression of cyclinDl protein was 13. 33% and 55. 13% . The abnormal expression of p - catenin protein and the positive expression of E2F - 1 , cyclinDl protein were related with the pathological grade of the colorectal cancer, clinical pathological staging and lymph node metastasis ( P <0. 05 ). In colorectal carcinoma, p -catenin and cyclinDl were positively correlated ( P < 0. 05 ); there was no significant correlation between p -catenin and E2F - 1 ( P > 0. 05 ), but E2F - 1 and cyclinDl were significantly correlated ( P < 0. 05 ). Conclusion The expression of E2F - 1, p -catenin and cyclinDl is related with the occurrence, development and metastasis of colorectal cancer and it can be the prognosis of the biological behavior of colorectal cancer.
