米力农对家兔顿抑心肌治疗效果的研究

     

摘要

目的:探讨磷酸二酯酶抑制剂米力农对家兔顿抑心肌的治疗效果。方法将新西兰家兔40只通过简单随机抽样为对照组、完全缺血组、心肌顿抑组和米力农组,各10只。对照组:不阻断家兔冠状动脉;缺血组:完全阻断家兔冠状动脉并不再开放;心肌顿抑组:阻断家兔冠状动脉30 min,开放60 min;米力农组:通过结扎冠状动脉左旋支30 min,开放60 min建立心肌顿抑模型。通过检测不同时间点(实验30 min,60 min,90 min)的肌钙蛋白指标变化及病理改变了解米力农对心肌细胞的影响,通过细胞凋亡的流式实验检测家兔心肌凋亡情况。结果对照组cTnI在整个实验过程中无明显变化[(45±7),(46±10),(45±7) mg/L]。缺血组[(279±5),(361±23),(482±34) mg/L]及心肌顿抑组[(281±3),(377±20),(494±26) mg/L]较对照组 cTnI 显著增高,差异有统计学意义(P<0.05)。米力农组:实验60 min、90 min cTnI较实验30 min时逐渐降低[(88±6) mg/L、(50±6) mg/L比(119±6) mg/L](P <0.05),心肌顿抑组和缺血组凋亡率和坏死率显著高于对照组[(36.2±8.1)%,(27.6±3.7)%比(21.0±2.4)%;(10.4±3.1)%,(17.6±3.8)%比(3.0±0.5)%](P<0.05)。心肌顿抑组凋亡率高于缺血组(P <0.05),而坏死率显著低于缺血组(P <0.05)。米力农组心肌细胞坏死率[(4.6±0.9)%]和凋亡率[(23.0±3.7)%]均显著低于心肌顿抑组(P<0.05),心肌细胞坏死率低于缺血组(P<0.05)。结论米力农能够减少家兔缺血/再灌注损伤所诱导的心肌细胞凋亡。%Objective To investigate the effects of milrinone on myocardial stunning of rabbits . Methods Forty New Zealand rabbits were divided into 4 groups randomly:control group,ischemia group, myocardial stunning group,milrinone group,10 rabbits each.In the control group:coronary artery not blocked in rabbits;ischemia group:rabbit coronary artery was completely blocked and was no longer open;myocardi-al stunning group:rabbits coronary artery blocked 30 min,open 60 min;milrinone group:ligation of left coro-nary artery for 30 min, open 60 min to establish myocardial stunning model.cTnI at different time points (experiment 30,60,90 min) were monitored to understand heart function changes and milrinone effect on cardiocytes.Apoptotic cardiomyocyte was detected flow cytometry of apoptosis method .Results The cTnI of the control group had no obvious change in the process of the whole experiment [ ( 45 ±7 ) , ( 46 ±10 ) , (45 ±7) mg/L].The cTnI of the ischemia group [(279 ±5),(361 ±23),(482 ±34) mg/L] and the my-ocardial stunning group [(281 ±3),(77 ±20),(494 ±26) mg/L] was significantly higher than the control group,the differences were statistically significant (P<0.05).Milrinone group:cTnI at experiment 60 min, and 90 min was lower than at 30 min[(88 ±6) mg/L,(50 ±6) mg/Lvs(119 ±6) mg/L,P<0.05].Myo-cardial apoptosis rate and myocardial necrosis rate of myocardial stunning group and ischemia group were sig-nificantly higher than control group[(36.2 ±8.1)%, (27.6 ±3.7)% vs (21.0 ±2.4)%;(10.4 ± 3.1)%,(17.6 ±3.8)% vs (3.0 ±0.5)%] (P<0.05).Myocardial apoptosis rate of myocardial stunning group was significantly higher than the ischemia group(P<0.05),and myocardial necrosis rate was signifi-cantly lower than the ischemia group(P<0.05).The apoptosis rate[(4.6 ±0.9)%] and the necrosis rate [(23.0 ±3.7)%] of milrinone group were significantly lower than those in myocardial stunning group(P<0.05),the necrosis rate was significantly lower than that in ischemia group(P<0.05).Conclusion Milri-none can decrease cardiomyocyte apoptosis induced by ischemia/reperfusion injury in rabbits.
