MMP-2在肾动脉狭窄合并动脉粥样硬化大鼠动脉斑块中的表达

             

摘要

目的 探讨明胶酶(MMP-2)在肾动脉狭窄合并动脉粥样硬化大鼠动脉斑块中的表达及意义.方法 建立一肾一夹(One Kidney One Clip,1K1C)组及二肾一夹(Two Kidney One Clip,2K1C)组肾动脉狭窄合并动脉粥样硬化的大鼠动物模型,检测各组大鼠肾素活性(PRA)及MMP-2在各组大鼠动脉斑块中的表达,并进行统计学分析.结果 1K1C及2K1C组PRA均有所升高,2K1C组与阴性对照组比较有统计学意义(P<0.05),免疫组化显示手术组大鼠主动脉弓粥样斑块处MMP-2的表达显著高于对照组(P<0.05),并且MMP-2在2K1C组中的表达明显高于1K1C组(P<0.05).结论 动脉粥样硬化大鼠动脉斑块中有MMP-2的高表达,并且RAS系统可能加速动脉粥样斑块的形成.%Objective To study the expression of MMP-2 in aortic plaque area of rats with renal artery stenosis and atherosclerosis. Methods Animal model establishment: 26 male SD rats were randomly divided into One Kidney One Clip (1K1C) group, Two Kidney One Clip (2K1C) group and control group. Renal artery stenosis was caused by surgery both in 1K1C group and 2K1C group. All of the rats were injected VitD3 500,000 IU/kg and feeded by high-fat for 10 weeks to establish the model of renal artery stenosis and atherosclerosis. Blood collection from heart after fasting and forbidden to drink for 8h, then detected the plasma renin radioimmunoassay activity (PRA) by automatic biochemical analyzer; using immunohistochemistry to detect the expression of MMP-2 in aortic arch plaque of experimental rats, comparing the average optical density values of each groups. Results Compare with the negative control group, PRA were increased significantly in 2K1C group (P<0. 05). Compared with negative control group, MMP-2 were all increased in the 1K1C and 2K1C group (P<0. 05) ,and compared with 1K1C group, MMP-2 increased significantly in 2K1C group(P<0. 05). Conclusion The high expression of MMP-2 can be detected in rats with renal artery stenosis and atherosclerosis, and the RAS system may accelerate atherogenesis progress.
