目的:探讨磷脂酰肌醇3激酶(PI3K)抑制剂LY294002对高糖培养的大鼠肾小球足细胞snail表达的影响,为进一步研究糖尿病肾病足细胞损伤的机制提供依据。方法:体外培养大鼠原代肾小球足细胞,经细胞免疫荧光法鉴定。足细胞分化完全后同步化,分为正常糖组、甘露醇对照组、高糖组、高糖+LY294002(2 mg/L)组,培养上述细胞48 h后观察细胞形态,Western blot法半定量检测snail的表达。结果:高糖可导致足细胞足突消失,细胞体积变大,snail表达增强(P<0.05)。与高糖组相比,高糖+LY294002(2 mg/L)组可部分抑制高糖导致的snail表达增强(P<0.05)。结论:PI3K抑制剂可部分阻断高糖导致的足细胞snail表达。%Objective:To evaluate the effect of the blocker of PI3K LY294002 on the expression of snail in high glucose-induced podocyte cell.Method:The glomeruli were isolated and gathered for primary culture. The cells were randomized into normal glucose(NG) group,high glucose(HG) group,mannitol control(MG) group,HG+LY294002(2 mg/L)group. The culture was continued for 48 h.The expression of snail was observed with Western blot methods.Result:Compared with NG group,the podocytic-process retraction and partial effacement and intercellular junction losing appeared in HG group,the expression of snail was increased(P<0.05). Compared with HG group,there was a down-regulation of the expression of snail(P<0.05).Conclusion:PI3K inhibitor protects podocytes by partly inhibiting the expression of snail.
