Objective] To explore the mechanism of Wnt/β-catenin signaling pathway antagonist protein Dickkopf-1(DKK1) in regulating bone formation and metabolism. [Method]By referencing the articles in recent 10 years in CNKI, VIP and PUBMED, summarizing and analysing the mechanism of DKK1 in bone development and metabolism related diseases.[Result] DKK1,a soluble inhibitor of Wnt/β-catenin signaling ,plays a crucial role in skeletal system development and bone pathologies. [Conclusion] Research shows that DKK1 influencing bone development through canonical Wnt/β-catenin signaling pathway, and neutralizing or inhibiting DKK1 may prove effective in treating bone related diseases. This review may provide new ideas for the treatment of metabolism related diseases and further research of bone formation and metabolism.%[目的]探讨Wnt/β-catenin信号通路拮抗蛋白Dickkopf-1(DKK1)在骨骼发育和骨代谢中的作用机制,为骨骼发育和骨代谢相关疾病的治疗奠定基础。[方法]查阅近10年内CNKI、维普数据库、PUBMED数据库中DKK1相关国内外文献,从DKK1对骨发育、骨代谢及相关疾病等方面分析总结其作用机制。[结果] DKK1是Wnt/β-catenin信号通路的可溶性抑制剂,它在骨骼发育和骨代谢的过程中都发挥着至关重要的作用。[结论] DKK1通过抑制经典Wnt/β-catenin通路影响骨发育,中和或抑制DKK1的表达对多种骨代谢疾病起到治疗作用,这也为骨骼发育和骨代谢机制研究及药物筛选提供新的思路。
