SDF-lα促进骨髓间充质干细胞在脑缺血/再灌注大鼠海马中的存活与分化

     

摘要

目的 观察基质细胞衍生因子1α(SDF-lα)对脑缺血/再灌注大鼠海马中移植的骨髓间充质干细胞(BMSCs)的作用.方法 培养成年大鼠BMSCs并用BrdU标记,"四血管阻断法"制备大鼠脑缺血/再灌注模型.造模成功大鼠复灌24 h后分为模型对照组、BMSC组和SDF-1α+BMSC组.后两组经尾静脉分别注射BMSCs或SDF-1α +BMSCs,模型对照组同法注射等体积生理盐水.分别于注射后存活1、3、7天和1个月时,应用免疫荧光化学法检测大鼠海马区域BrdU标记阳性细胞的表达情况.结果 免疫荧光染色显示:①模型对照组大鼠在移植不同时间点海马结构中均未见BrdU标记的阳性细胞,但BMSC组和SDF-1α+BMSC组的BrdU标记阳性细胞数随移植时间延长而逐渐增多,且各时间点间差异均有统计学意义(P<0.05);②SDF-1α+BMSC组移植后1、3、7天BrdU阳性细胞数均显著高于相应时间点的BMSC组(P<0.05),其中7天时的差异最显著(P<0.01),但1个月时两者差异无统计学意义(P>0.05).结论 SDF-1α可显著促进BMSCs在脑缺血/再灌注大鼠海马中的存活与分化,提示SDF-1α在BMSCs移植治疗缺血性脑损伤过程中可起到促进作用.%Objective To investigate the effect of bone marrow mesenchymal stem cells ( BMSCs) mediated by SDF - la at different time points after transplanted BMSCs in rat hippocampus subjected to cerebral ischemia /reperfusion (I/ R). Methods Cultured BMSCs were labeled by the BrdU and " four vessel occlusion " method were used to estahlish cerebral I/R model. The models were divided into three groups : control group, BMSCs group and SDF - lα + BMSCs group. After reperfusion for 24 h, the normal saline , BMSCs and SDF - lα + BMSCs were transplanted into the I/R model by the tail vein respectively , and immunofluorescence determination was employed for examining the BMSCs of migra -ting to hippocampus at different time points of transplanting the BMSCs . Results Immunofluorescence staining showed that there was no positive cell of BMSCs labeled BrdU ( BrdU+ - BMSCs ) in the hippocampus of the control group ; BrdU+ -BMSCs were found in the hippocampus of BMSCs group and SDF - lα + BMSCs group at transplanting different time points, and the number of positive cells were increased following transplanting time ; and the number of positive cells were significant difference at transplanting 3 d, 7 d and 1 M hippocampus compared with 1 d (P <0. 05). Compared with BMSCs group, the number of positive cell had significant difference at transplanted 7 d of SDF - lα + BMSCs group in hippocampus (P < 0.05 ) , but there was no statistical difference at 1 M group ( P > 0.05 ). Conclusion The SDF -lα can promote the survival and differentiation of BMSCs , and these results suggest that the SDF - la plays a stimulative role during the BMSCs treats the ischemic cerebral injury .
