男性痛风患者与TLR4基因rs10759932(C/T)和rs1927906(C/T)位点单核苷酸多态性的研究

         

摘要

Objective To investigate the association of Toll-like receptors (TLR) 4gene rs10759932 (C/T) and rs1927906 (C/T) polymorphisms with male gout.Methods A total of 103hospitalized patients (gout group) of the Department of Rheumatology in the First Affiliated Hospital of Xinjiang Medical University and Xinjiang Uygur Autonomous Region People′s Hospital versus 103healthy subjects (control group) were collected.The age portion of the two groups was measured, and the biochemical indicators, height, body mass and blood pressure were measured.The TLR4gene rs10759932 (C/T) and rs1927906 (C/T) loci were detected by PCR.Results The genotype distribution of the two loci was consistent with the Hardy-Weinberg genetic equilibrium law (P>0.05) , and the population was representative.The frequency of CC, CT, and TT genotypes in the rs10759932locus were 10%, 48%, and 42%respectively in the gout group, and 6.1%, 34.7%, and 59.2%respectively in the control group.The CC and CT genotype cases were higher than the control group with statistical significance (P0.05).Conclusion The single nucleotide polymorphism of rs10759932locus is associated with the onset of gout in males.It is not considered that the single nucleotide polymorphism at rs1927906is associated with the onset of gout in men.%目的 探讨Toll样受体 (toll-like receptors, TLR) 4基因rs10759932 (C/T) 和rs1927906 (C/T) 位点多态性与男性痛风的相关性.方法 收集新疆医科大学第一附属医院、新疆维吾尔自治区人民医院风湿科痛风住院患者 (痛风组) 103例和同期健康体检者 (对照组) 103例, 对2组人群进行年龄配比, 测量生化指标、身高、体质量、血压.并采用PCR技术检测TLR4基因rs10759932 (C/T) 和rs1927906 (C/T) 位点基因型, 所有数据采用SPSS22.0进行统计分析.结果 两个基因位点的基因型分布符合Hardy-Weinberg遗传平衡定律 (P>0.05) , 群体具有代表性.rs10759932位点CC、CT、TT基因型在病例组的分布频率分别为10%, 48%和42%, 对照组中为6.1%, 34.7%和59.2%, CC和CT基因型病例组高于对照组, 差异有统计学意义 (P0.05) .结论 rs10759932位点单核苷酸多态性与男性痛风的发病有关, 尚不能认为rs1927906位点单核苷酸多态性和男性痛风发病有关.
