HPLC-RID 法测定不同批次刺糖中蔗果三糖含量

     

摘要

目的:建立测定刺糖中蔗果三糖(GF2)含量的高效液相色谱-示差折光检测法(HPLC-RID),测定10个不同批次刺糖中 GF2的含量。方法采用 YMC-Pack NH2色谱柱(250 mm×4.6 mmL.D.,S-5μm,12 nm)分离;流动相:乙腈-水=75∶25,流速:1.0 mL/min,进样量:30μL。结果GF2在0.808~1.616 mg/mL 浓度范围内线性关系良好,r =0.9994;精密度相对标准差为0.57%;低、中、高浓度加样回收率分别为97.8%、96.1%、104.3%,相对标准差分别为1.8%、1.8%、1.6%。10批次刺糖中 GF2的含量最高为454.57 mg/g,最低为115.90 mg/g。结论该方法精密度、准确度良好,适用刺糖中 GF2的定性及定量分析。%Objective To establish the method for the determination of GF2 in Alhagi-honey by HPLC-RID and to determine the content of GF2 from the 10 different batches of samples.Methods The separation was performed using YMC-Pack NH2 Column (250 mm×4.6 mmL.D.S-5 μm,12 nm)with acetonitrile-water (75∶25)as mobile phase at a flow rate of 1.0 mL/min.Results The GF2 showed there was a good linear relationship in the range of 0.808- 1.616 mg/mL,r =0.9994;The relative standard deviation of precision was 0.57%;The recovery rates of low-,middle-,and high-concentration of GF2 were 97.8%, 96.1% and 104.3%,respectively;the relative standard deviation were 1.8%,1.8% and 1.6%.The highest content of GF2 from the 10 batches of samples was 454.57 mg/g,and the lowest was 115.90 mg/g.Conclu-sion The method has been established with high precision and accuracy ,which could be used for the qualitative and quantitative analysis of GF2 from Alhagi-honey.
