小鼠灌胃乙酸建立胃溃疡模型的可行性研究

         

摘要

Objective To explore the feasibility of intragastric administration of acetic acid for inducing gastric ulcer model in mouse,and explore its reasonable modeling conditions.Methods Sixty mice were randomly divided into six groups according to the concentration of acetic acid:7.34 × 10-4mol/L group,8.74 × 10-5 mol/L group,1.75 × 10-5 mol/L group,8.74 × 10-6 mol/L group and 4.37 × 10-6 mol/L group and saline control group.Each group contained five males and five females.Acetic acid was intragastric administrated at the volume of 0.3 ml each time.After the model was established,the presence and degree of ulcer were observed with naked eyes and microscope.Results Superficial ulcer was found in all mice after 7.34 × 10-4 mol/L acetic acid for one time and 8.74 × 10-5 mol/L for three times.The intragastric administration of 1.75 × 10-5 mol/L acetic acid for 5 times resulted in superficial ulcer in 50% cells.Under microscope,typical peptic ulcer and the damage of both glands and mucosa were found after 8.74 × 10-6 mol/L for seven times.Only slight edema and congestion in mucosa was found after 4.30 × 10-6 mol/L acetic acid for seven times.Conclusion It's feasible to induce mouse gastric ulcer by intragastric administration of acetic acid.And the degree of gastric ulcer is related to the concentration of acetic acid.%目的 探讨乙酸灌胃法建立小鼠胃溃疡模型的可行性,并确立合理建模方案. 方法 60只昆明种小鼠随机分为6组,每组10只,雌雄各半分笼饲养,分别为7.34×10-4mol/L,8.74×10-5 mol/L,1.75×10-5 mol/L,8.74×10-6mol/L,4.30×10-6 mol/L乙酸溶液灌胃组以及生理盐水对照组,各组每次灌胃药量均为0.3ml.模型建立后对各组小鼠是否出现溃疡及其程度进行肉眼和光镜观察. 结果 灌胃7.34×10-4mol/L乙酸溶液1次,8.74×10-5 mol/L乙酸溶液3次后所有动物均出现浅表性溃疡,灌胃1.75×10-5 mol/L乙酸溶液5次后有50%个体出现浅表性溃疡,其余个体无明显病理改变,与对照组比较差异有统计学意义(P<0.01);灌胃8.74 × 10-6 mol/L乙酸溶液7次后,在光镜下可见典型的消化性溃疡结构,周边黏膜腺体正常结构消失.灌胃4.30×10-6 mol/L乙酸溶液7次后,胃黏膜仅发生轻度充血水肿,未见明显病理改变. 结论 乙酸灌胃法制作小鼠胃溃疡模型简便易行,溃疡程度与灌胃乙酸的浓度有关.
